Abstract
Fitopatogena gljiva Ophiostoma novo-ulmi uzročnik je druge pandemije izrazito destruktivne holandske bolesti brijesta. Vrsta O. novo-ulmi dijeli se na dvije podvrste: O. novo-ulmi ssp. novoulmi i O. novo-ulmi ssp. americana, između kojih postoji parcijalna reproduktivna barijera zbog čega je moguća njihova hibridizacija u prirodi. U ovom radu analizirani su izolati gljive O. novo-ulmi s područja Hrvatske. Za determinaciju podvrsta i njihovih hibrida primijenjena je PCR-RFLP analiza gena cu i col1. Kako bi se istražilo postoje li razlike u virulentnost izolata različitih podvrsta i hibrida primijenjena je metoda inokulacije izolata na plod jabuke (Malus x domestica 'Golden delicious') te je virulentnost procijenjena na temelju promjera nekrotične lezije nastale nakon 14 i 28 dana. Utvrđene su statistički značajne razlike u veličini promjera nekrotičnih lezija, osobito 28 dana nakon inokulacije, što ukazuje na postojanje značajnih razlika u virulentnosti pojedinih izolata gljive O. novo-ulmi. Međutim, virulentnost testiranih izolata nije se mogla dovesti u korelaciju s rezultatima molekularne identifikacije patogena. Ovo istraživanje predstavlja doprinos karakterizaciji uzročnika holandske bolesti brijesta u Hrvatskoj, a po prvi puta je opisano postojanje razlika u virulentnost izolata O. novo-ulmi s područja Hrvatske.
Abstract (english)
Phytopathogenic fungus Ophiostoma novo-ulmi is a casual agent of the second, extremely destructive, pandemic of Dutch elm disease. Species O. novo-ulmi is divided into two subspecies: O. novo-ulmi ssp. novo-ulmi and O. novo-ulmi ssp. americana. With only partial reproductive barrier between them, hybridization takes place in nature. In this study O. novo-ulmi isolates from Croatia were analyzed. Determination of subspecies and their hybrids was based on PCR-RFLP analysis of genes cu and col1. Virulence of isolates determined as different subspecies and hybrids was investigated by inoculation of isolates on apple fruit (Malus x domestica 'Golden delicious') and evaluated based on diameter size of necrotic lesion observed after 14 i 28 days. Statistically significant differences in diameter size of necrotic lesions were identified, especially after 28 days post inoculation. Those results indicate significant differences in virulence of individual O. novo-ulmi isolates. However, virulence of tested isolates was not in correlation with the results of molecular identification of the pathogen. This research represents contribution to characterization of casual agent of Dutch elm disease in Croatia. For the first time, the existence of differences in virulence of O. novo-ulmi isolates found in Croatia was indicated.
