Abstract
Male interferirajuće RNA (siRNA), RNA sa strukturom ukosnice (shRNA) i mikro RNA (miRNA) nekodirajuće su molekule koje imaju važnu ulogu u regulaciji ekspresije gena. Ove molekule se koriste kao posrednici u terapeutskom liječenju različitih bolesti kao što su infekcije i tumori. Sve tri molekule su dvolančane RNA molekule koje posredovanjem u procesu RNA interferencije rezultiraju utišavanjem gena komplementarnim sparivanjem s ciljnom mRNA molekulom. Ove se RNA molekule suočavaju sa sličnim preprekama tijekom RNAi: slaba stabilnost in vivo, poteškoće u isporuci molekule zbog barijera te neželjeni efekti koji ne rezultiraju utišavanjem gena. Najveći napredak u rješavanju tih nedostataka leži u dizajniranju sintetičkih molekula s poboljšanim karakteristikama, kemijskim modifikacijama te vektorima za isporuku molekula u ciljne stanice. Rezultati RNAi pokazuju da će terapije bolesti ovim metodama imati i široku primjenu u budućnosti
Abstract (english)
Small interfering RNA (siRNA), short hairpin RNA (shRNA) and micro RNA (miRNA) are non-coding molecules that play an important role in regulation of gene expression. These molecules are used as intermediates in the therapeutic treatment of various diseases such as infections and tumours. All three molecules are double-stranded RNA molecules that by mediating in the RNA interference process result in the suppression of the gene by complementary coupling with the target mRNA molecule. These RNA molecules face similar obstacles during RNAi: weak in vivo stability, difficulty in delivery of the molecule due to barrier, and unwanted effects that do not result in the suppression of the gene.
