Abstract
Stanične membrane su ključne za održavanje normalnog metabolizma stanice, reguliraju prolaz tvari između stanice i okolnog medija. Lijekovi nakon unošenja u organizam moraju proći kroz membranu radi dolaska do ciljanog mjesta i djelovanja. Određene vrste lijekova mogu svojim djelovanjem i sastavom oštetiti membranu, čime ona mijenja ili gubi svoju funkciju. Lijekovi mogu utjecati na propusnost, fluidnost i zakrivljenost membrane, dovesti do grupiranja lipida, te ometati koncentracijski gradijent stanične membrane. Neki se lijekovi vežu za membranske proteine i receptore te zamijenjuju lipide koji grade membranski sloj, dok neki mogu inhibirati ili aktivirati ionske kanale kroz interferenciju sa konformacijskim promjenama. Promjena funkcije membrane djelovanjem lijekova treba biti i u fokusu istraživanja novih antibiotika, zbog činjenice da su brojne bakterije razvile opornost prema dosadašnjim lijekovima.
Abstract (english)
Cell membranes are important for maintaining normal cell metabolism, they regulate the transport of substances between the cells and the surrounding medium. Medicines, when introduced into the body, must pass through membranes in order to reach the target site and act. Certain types of medicines can damage the membrane through their action and composition, causing it to change or lose its function. Medicines can affect the permeability, fluidity and curvature of the membrane, lead to the grouping of lipids, and interfere with the concentration gradient of the cell membrane. Some medicines bind to membrane proteins and receptors and replace the lipids that build the membrane layer, while others can inhibit or activate ion channels through interference with the conformational changes. Changed membrane function due to the action of medicines should also be the focus of research of new antibiotics, due to the fact that numerous bacteria have developed resistance to existing medicines
