Abstract
Nakupljanje i distribucija lijekova u kostima su procesi koji ovise o različitim čimbenicima, kao što su građa kosti, fizikalno-kemijska svojstva lijeka i metabolička aktivnost koštanih stanica. Razumijevanje mehanizama nakupljanja i distribucije lijeka u kostima važno je za optimizaciju terapije lijekovima i minimiziranje nuspojava. Isporuka lijeka trebala bi biti što preciznija kako bi se postigla maksimalna učinkovitost i sigurnost tretmana. Lijek može do kosti biti dopremljen u kompleksu s nosačima lijekova i agensima za pronalazak kosti, što je efikasniji i manje invazivan način dopreme. Visoki afinitet određenih lijekova, poput bisfosfonata, za koštani matriks može dovesti do njihove akumulacije i omogućiti trajni terapijski učinak. S druge strane, često propisivani lijekovi koji nisu namijenjeni za tretman bolesti kostiju mogu rezultirati nuspojavama izraženim na ljudskom skeletu
Abstract (english)
Accumulation and distribution of medicines in bones are processes that depend on various factors, such as bone structure, physicochemical properties of the drug, and metabolic activity of bone cells. Understanding the mechanisms of drug accumulation and distribution in bones is important for optimizing drug therapy and minimizing side effects. The delivery of the medicine should be as precise as possible in order to achieve maximum efficiency and safety of the treatment. The medicine can be delivered to the bone in a complex with drug carriers and bone-seeking agents, which is a more efficient and less invasive way of delivery. The high affinity of certain drugs, such as bisphosphonates, for the bone matrix can lead to their accumulation and enable a lasting therapeutic effect. On the other hand, frequently prescribed drugs that are not intended for the treatment of bone diseases can result in side effects expressed on the human skeleton.
