Abstract
Tehnologija CRISPR/Cas9 je alat genetičkog inženjerstva. Sam sustav je otkriven u imunosnom sustavu bakterija te je prilagođen za primjenu u biljnim i životinjskim stanicama uključujući i čovijeka. Şustav se sastoji od dvije domene, preoteina Cas9 koji sadrži NHN i RuvC domene za iskrivljenje ciljane DNA i gRNA koja dovodi protein Cas9 do ciljanog mjesta na DNA. Sustav izaziva dvolančane lomove na DNA koji se dalje popravlja mehanizmom prirodno urođenom stanici. Tehnologija je značajna i privlači puno pozornosti zbog svoje jednostavnosti, široke mogućnosti primjene i niske cijene. Ima širok spektar mogućih primjena kao što su mijenjanje genoma biljnih i životinjskih stanica, liječenje bolesti, istraživanja pojedinih lokusa genoma. Jedan od problema primjene tehnologije se odnosi na modifikacije ljudskih reproduktivnih stanica čije osobine se prenose kroz generacije te strah od zloupotrebe tehnologije.
Abstract (english)
CRISPR/Cas9 technology is one of the tools in genetic engineering. System is dicovered as a part of bacterial imunosystem and its developed for use in plant and animal cells including humans. System contains two parts, Cas9 protein wich contains NHN and RuvC domains for targeted DNA cleavage and gRNA wich leads Cas9 protein to the targeted DNA locus. System causes DSB wich is then repaired by native cells mechanism. The tehnology is significant and atracts a lot of attention because of its simplicity, wide possibility of uses and low price. It has wide spectrum of possible uses like a genome editing, in plant and animal cells, curing ilnesses, researching unique genom locuses. One of the issues of tehnology applications refers on the modifications of the human reproduction cells whose genetic material is transferred through generations and the fear of misuse of tehnology.
