Abstract
Dvoklasićasta kostrika (Brachypodium distachyon (L.) P. Beauv.) je jednogodišnja biljka koja pripada porodici Poaceae. Vrlo se jednostavno uzgaja, ima kratko generacijsko vrijeme, malen i kompaktan genom ( ~ 300 Mpb) te je filogenetski srodstvena s agronomski važnim kulturama kao što su pšenica (Triticum aestivum L.), soja (Glycine max L. Merr.), riža (Oryza sativa L.) i zob (Avena sativa L.) što ju čini prihvatljivim modelom u brojnim istraživanjima. Najčešće tehnike transformacije uključuju bombardiranje česticama (particle bombardment) te transformacija s bakterijom roda Agrobacterium. Za provođenje tih tehnika stvara se kultura kalusa od izoliranih embrija na različitim medijima. U ovom je radu optimizirane su metode stvaranja kulture kalusa iz embrija na različitim medijima.
Abstract (english)
Brachypodium distachyon (l.) P. Beauv. s an annual plant form Poaceae family. It is easily grown and has short generation period. Its genome is short and compact (~ 300 Mbp) and it is phylogenetically related to important species in agronomy such as wheat (Triticum aestivum L.), soy (Glycine max L. Merr.), rice (Oryza sativa L.) and oats (Avena sativa L.) which makes her acceptable as model in numerous research. Particle bombardment and Agrobacterium transformation are the most common techniques used on Brachypodium distachyon. Callus culture from isolated embryos is created for implementing those techniques on different kinds of medium. In this work methods of creating embryo culture are optimized using different growth medium.
